Brendon Little was born and raised in the United States. He graduated from Conestoga High School in 2015, where he began to develop his skills as a baseball player. Instead, he chose to pursue collegiate baseball at the University of North Carolina.

He began the 2024 season with the Triple-A Buffalo Bisons before being called up to the Blue Jays on June 1, 2024, when he joined the active roster due to an injury to teammate Jordan Romano.
